Organoids are self-organized three dimensional tissue cultures mimicking in vivo organs. They have great potential capable of recapitulating tissue generation and function in basic and translational research. For clinical application such as drug screening, efficacy evaluation, toxicity test, pharmacokinetics profiling, tissue regeneration, and transplantation, organoids provide ex vivo model as well as platform. In this lecture, recent research progress on organoid generation, application, and functional mechanism will be handled. Also establishment of robust intestinal organoids from embryonic stem cell and adult stem cell will be introduced. The evaluation of successful establishment of intestinal organoids made by transcriptome and epigenome analysis will be provided. The gene regulation network and differentiation/proliferation pathway during organoid formation show an insight on the potential and value of organoids as a disease modeling.-
