- Abstract
- Surgical practice for treatment of small renal masses has shifted from radical nephrectomy towards partial nephrectomy, providing opportunities for removal of tumors while preserving normal renal mass and renal function. This study aimed to develop kidney-derived extracellular matrix bioink and a construct that can be surgically sutured to the damaged kidney to provide cues for regeneration of the kidney tissue. As a base material for the construct, porcine kidney decellularization was performed along with its quantification of glycosoaminoglycans (GAGs), collagen, and DNA. Immunohistochemical staining of the decelluarized tissues showed a high preservation of the major extracellular matrix (ECM) components. Rheological analysis yielded the printability of the kidney decellularized extracellular matrix (kdECM) bioinks at 1%, 2%, and 3%. However, difference in viability were represented among the organoids printed in different bioink concentrations. Accordingly, results from this study on the kidney decellularized ECM based construct can be referred to provide a step closer to developing a kidney tissue mimicking construct that can be applied for regenerative approaches.
