dc.description.abstractVaccinia-relatedkinases(VRKs)aremultifacetedserine/threoninekinasesthatplayessentialrolesinvariousaspectsofcellsignaling,cellcycleprogression,apoptosis,andneuronaldevelopmentanddifferentiation.However,theneuronalfunctionofVRK3isstillunknowninspiteofitsetiologicalpotentialinhumanautismspectrumdisorder(ASD).Here,wereportthatVRK3-deficientmiceexhibitedtypicalsymptomsofautism-likebehaviorincludinghyperactivity,stereotypedbehaviors,reducedsocialinteraction,andimpairedcontextdependentspatialmemory.SignificantdecreaseindendriticspinenumberandarborizationwereidentifiedinthehippocampusCA1ofVRK3-deficientmice.ThesemicealsoexhibitedareducedrectificationofAMPAreceptor-mediatedcurrentandchangesinexpressionofsynapticandsignalingproteinsincludingTrkB,ArcandCaMKIIα.Notably,TrkBstimulationwith7,8-dihydroxyflavone(7,8-DHF)reversedthealteredsynapticstructureandfunction,andsuccessfullyrestoredautism-likebehaviorinVRK3-deficientmice.TheseresultsrevealthatVRK3playsacriticalroleinneurodevelopmentaldisordersandsuggestapotentialtherapeuticstrategyforASD.-
