- Abstract
- The liver is the largest internal organ that regulates critical biochemical functions of body. Nowadays, alternatives to donor organ for end stage liver diseases and in vitro model for predicting hepatotoxicity or biotransformation of drug candidates have been studied. However, due to the complexity of native liver tissue, the research progresses are still at a fundamental stage. Recently, three-dimensional (3D) cell printing technology has emerged as a promising approach to overcome the challenge of the deliberate positioning of the cells and biomaterials; yet, there are few research on the development of bioinks suitable for liver construct fabrication. Therefore, we developed a liver decellularized extracellular matrix (LdECM) derived bioink to mimic the biochemical environments of the native liver tissue. Firstly, native liver tissue was decellularized and solubilized into printable bioink. It showed suitable and adjustable mechanical properties for 3D cell printing, and the printing parameters were studied with the liver dECM bioink to verify the versatility and fidelity of the printing process. In addition, in the 3D cell printed constructs using LdECM bioink, the hepatic differentiation potential of mesenchymal stem cell and enhanced hepatic functions of HepG2 were observed. Consequently, the results demonstrated that the LdECM bioink would be one of the promising candidates for fabricating liver constructs using 3D cell printing technology.
